What significance test?
Method used depends on rarity of incidence of interest
If “leukemia rare” then Poisson probabilities are appropriate
If “fairly common” then Poisson or binomial may be appropriate
If “very common” then binomial or bootstrap an excess statistic may be appropriate
If in doubt then use a Monte Carlo Significance test
